摘要
色氨酸是人和动物必需的8种氨基酸之一。由于L-色氨酸特殊的结构和生物学功能,它在人和动物的生长发育及新陈代谢中有着至关重要的作用,因而被广泛应用于食品、饲料添加剂、医药、农业及环境监测等方面。随着人们对色氨酸的用途越来越了解,色氨酸的市场会迅速增加,前景乐观。L-色氨酸工业属于现代发酵工程,本设计是L-色氨酸的提取车间设计。以葡萄糖为原料,以大肠杆菌为生产菌株,采用发酵法生产L-色氨酸,经膜分离、离交、脱色、超滤、真空浓缩、结晶、干燥等工艺提取L-色氨酸。设计年产纯度为98.5%以上的结晶L-色氨酸4000吨,产酸为3.8%,装液量/罐容为75%,L-色氨酸总提取收率为81%,发酵周期34 h,每年生产320天。本设计就L-色氨酸的提取工艺流程、物料衡算、热量衡算、设备的选型、工厂布置、车间设计、水、电、气、汽以及辅助设备、三废处理及综合利用等方面对L-色氨酸提取车间进行设计。整个设计考虑了目前的经济状况和今后的发展需求,从节能角度选择设备且生产流程尽量保证机械化、自动化,同时提取做到技术先进,产品质量合格,车间布局规范合理。

Abstract
Tryptophan is one of the eight kinds of essential amino acids for humans and animals. Due to the special structure and biological function of L- tryptophan, has a very important role in the growth and development of humans and animals as well as metabolic process, it is widely used in food and feed additives industry, pharmaceuticals, agriculture and environmental monitoring. As people become more and more aware of the use of tryptophan, tryptophan market will increase rapidly, the outlook is optimistic. L-trptophan industry belongs to the modern fermentation engineering, this work is a L-trptophan extraction workshop design, with oral glucose as raw material, using Escherichia coli as production strain, the fermentation production of L-trptophan, by membrane separation, ion exchange, decolorization, ultrafiltration, vacuum concentration, crystallization, drying etc. steps extract L-trptophan. Design of purity of crystallization of more than 98.5% L-trptophan 4000 tons, acid production rate was 3.8%, the volume of liquid / tank capacity is 75%, the total yield of L-trptophan is 81%, the fermentation period is 34 h, 320 days for every year. According to L-trptophan extraction design of process flow, plant material and heat balance calculation, equipment selection, process layout, workshop design, water, electricity, gas, steam and auxiliary equipment and other aspects for L-trptophan extraction workshop design. This design takes full account of current economic situation and future development needs, chooses equipments from the perspective of energy-efficient, production processes to maximize mechanization, automation level, while seeking to technically advanced, reliable quality, reasonable and normal layout.

第一章前言

第一节L-色氨酸的理化性质

色氨酸是人和动物生命活动中八种必需氨基酸之一,在人和动物的生长发育、新陈代谢起着重要作用,是第二必需氨基酸。在1902年,Hokinst第一次从酪蛋白中分离获得色氨酸。其英文名为L-Tryptophan(L-Trp),学名为β-吲哚基丙氨酸,化学名称是L-2-氨基-3-吲哚基丙酸,别名为L-氨基吲哚丙酸、L-胰化蛋白氨基酸。分子式为C11H12O2N2,L-色氨酸的相对分子质量为204.21。
